Platelets last from 7-10 days. It is normal for PLT to vary by as much as 100K in a single day based on what is going on in the body. The body is constantly making new platelets with the volume normally being driven by the release of thrombopoietin (TPO), a ligand that is the "on switch" for making platelets. What is happening due to the MPN driver mutation is that the body is making platelets in the absence of TPO. In essence, the "on switch" in the mutated blood progenitor cells is always "on."

You PLT count can vary from day to day. That is why it is the trend over time that matters more than a single lab read. My PLT used to routinely vary by as much as 200K when I was not using cytoreductive treatment. The 500s - 700s was my baseline.
